Mechanism of Action
CJC-1295 without DAC (also known as Modified GRF 1-29) is a 29-amino acid growth hormone releasing hormone (GHRH) analogue consisting of the first 29 amino acids of endogenous GHRH with four amino acid substitutions: D-Ala at position 2 (for DPP-4 resistance), Gln at position 8, Leu at position 27, and Ala at position 28 (for enhanced stability and receptor affinity). Without the Drug Affinity Complex (DAC) moiety present in the DAC variant, CJC-1295 without DAC has a shorter half-life of approximately 30 minutes — closer to the physiological pulsatile pattern of endogenous GHRH. Upon binding the GHRH receptor (GHRHR) on pituitary somatotropes, CJC-1295 without DAC activates Galphas/cAMP/PKA signalling, stimulating growth hormone synthesis and release. The shorter half-life makes this variant suitable for research models that require pulsatile GH release signalling — the pattern that most closely mimics physiological GH secretion, where GH is released in distinct pulses driven by the interplay of GHRH stimulation and somatostatin inhibition. This contrasts with the sustained (basal) GH elevation produced by the DAC variant, making CJC-1295 without DAC the preferred research tool for studying physiological GH release patterns and pulsatile GHRH receptor signalling.
