Mechanism of Action

GHRP-2 (Growth Hormone Releasing Peptide-2, also known as Pralmorelin) is a synthetic hexapeptide (D-Ala-D-betaNal-Ala-Trp-D-Phe-Lys-NH2) that acts as a potent agonist of the ghrelin receptor (GHS-R1a, growth hormone secretagogue receptor). This distinct pathway enables GHRP-2 to stimulate GH release through a mechanism complementary to GHRH, and the two pathways produce synergistic GH release when combined. Beyond GH release, GHRP-2's ghrelin receptor agonism produces additional effects: ghrelin is the 'hunger hormone' and GHRP-2 stimulates appetite through hypothalamic ghrelin receptor activation. The peptide also modulates energy homeostasis signalling and may influence sleep architecture through central ghrelin receptor activity. GHRP-2 is studied alongside GHRH analogues (such as CJC-1295 or sermorelin) for combined GHRH-ghrelin receptor pathway research.
