Mechanism of Action

Kisspeptin-10 (also known as metastin-45-54) is a decapeptide derived from the kisspeptin protein encoded by the KISS1 gene. Kisspeptin-10 is a potent agonist of the kisspeptin receptor (KISS1R, also known as GPR54) — a class A G protein-coupled receptor expressed on GnRH neurons in the hypothalamus. The kisspeptin-kisspeptin receptor system is the primary upstream regulator of GnRH release: kisspeptin neurons in the arcuate and anteroventral periventricular (AVPV) nuclei project to GnRH neuron terminals, and kisspeptin binding to KISS1R on GnRH neurons activates Galphaq/PLCbeta signalling, producing calcium mobilisation and stimulating GnRH release into the hypophyseal portal system. This kisspeptin-GnRH-gonadotrope (LH/FSH) cascade makes kisspeptin-10 a powerful research tool for studying the upstream regulation of the hypothalamic-pituitary-gonadal (HPG) axis. Kisspeptin-10 is particularly significant in puberty onset research: KISS1R mutations cause hypogonadotropic hypogonadism (failure of puberty), establishing kisspeptin signalling as essential for reproductive axis activation. Kisspeptin-10's short sequence (10 amino acids) makes it more suitable for research applications than the longer kisspeptin-54, while retaining full KISS1R agonist activity.
