Mechanism of Action
Kisspeptin-54 (also known as metastin-68-121) is a 54-amino acid peptide fragment derived from the kisspeptin protein encoded by the KISS1 gene. Like kisspeptin-10, kisspeptin-54 is an agonist of the kisspeptin receptor (KISS1R/GPR54), activating Galphaq/PLCbeta signalling on GnRH neurons to stimulate GnRH release. The 54-amino acid kisspeptin-54 is the longer naturally occurring form, and is studied alongside the shorter kisspeptin-10 for comparative KISS1R agonism research. The key distinction between kisspeptin-54 and kisspeptin-10 is duration of action: kisspeptin-54's longer sequence provides additional structural features that confer extended receptor interaction and sustained signalling compared to the shorter kisspeptin-10, which has a more transient effect. This makes kisspeptin-54 suitable for research models requiring sustained KISS1R activation and prolonged GnRH release stimulation, while kisspeptin-10 is preferred for acute, transient receptor activation studies. Both peptides share the same C-terminal decapeptide region (kisspeptin-10 sequence) that contains the KISS1R-binding domain, but kisspeptin-54's additional N-terminal residues influence pharmacokinetics and receptor interaction kinetics.
