Mechanism of Action
Sermorelin is a synthetic peptide consisting of the first 29 amino acids of naturally occurring growth hormone releasing hormone (GHRH) — the minimal sequence retaining full biological activity at the GHRH receptor (GHRHR). As a GHRH analogue, sermorelin binds GHRHR on pituitary somatotropes and activates Galphas/cAMP/PKA signalling, stimulating both the synthesis and pulsatile release of growth hormone. The pulsatile pattern of GH release induced by sermorelin is physiologically significant: GH is naturally secreted in distinct pulses, with the pattern (pulse amplitude and frequency) carrying biological information distinct from sustained GH elevation. Sermorelin's short half-life (~10-12 minutes) produces a transient GHRH receptor activation that mimics endogenous pulsatile GHRH signalling, making it suitable for research models that require physiological GH release patterns. Sermorelin is studied alongside other GHRH analogues (CJC-1295 with and without DAC, tesamorelin) for comparative GHRH receptor pharmacology: while all activate the same receptor, they differ in half-life (from minutes to days) and thus in the temporal pattern of GH release they produce — pulsatile (sermorelin, CJC-1295 without DAC) versus sustained (CJC-1295 with DAC, tesamorelin).
